Apparent F concentrations of 4, 10, 25 and 62.5 mg/L were used to assess their effect on juvenile Siberian sturgeon comparing to the control which fluorine concentration in the water was 0.06 mg/L. Three replications (namely 3 tanks) were recorded for each F concentration and also for the controls. The water used for controls and other treatments for dilution was filtered tap water pre-aerated for more than 3 days. Stock solutions of 1000 mg F–/L were prepared from analytical reagent grade NaF in double distilled water and then diluted with water mentioned above to obtain the desired concentrations. Each tank contained 80 L solution. No addition of any chemical was made to the controls. The medium was mixed well after the addition of NaF for fluoride treatments. Each solution was replaced daily with 4/5 of the total solution by pumping the solution in to the rearing tank. The water quality and feeding was mentioned above.The exposure period was 90 days for all the treatments. Aim to get the data before fluoride exposure started, 2 fish were chosen randomly from each tank obtaining a total of 30 fish to do blood sampling and get tissues one day before fluoride exposure started. Then, at every 30 d interval, 6 fish from each tank, namely, 18 fish from each treatment were sampled to get blood. And, among these fish, 2 fish were randomly chosen from each tank,Tissues including bone, cartilage, skin, liver, gill, gut, pylorus, and notochord were sampled to determine fluorine concentration. In this study bone included dorsal scute and pectoral fin spine. Cartilage mainly composed of the head except the brain, skin on the head, eyes, skull roofing bones, gills and all other soft parts. Skin were gotten rid of muscle and scutes. Gill contains gill arch and the soft fillets. Gut was rinsed in distilled water. Notochord was the soft part inside the back cartilage. Besides parts of the tissues separated for histopathological analysis, all the tissues were stored at -80℃ till fluorine concentration determination. Small pieces of the liver, kidney, and gill tissues were fixed in Bouin’s solution for 24 h and then kept in 70% ethanol for histopathological examination.

Conjugate peak refinement (CPR) calculations
Calculations were performed using the CPR algorithm in the CHARMM molecular dynamics program.
Polar and aromatic hydrogens were included explicitly, and a non-bond cutoff of 8.5Å was used to minimize calculation of long-range interactions.
Since the CPR method does not allow the inclusion of explicit solvent, a distance-dependent dielectric was used.
The starting structures of deoxy HbA and oxy HbA were obtained from the Protein Data Bank (PDB) .
Each hemoglobin structure was energy minimized to alleviate bad atomic contacts. Cross-linkers were built using the modeling program QUANTA (Molecular Simulations, Inc.) and energy minimized.
The cross-linker was positioned in the central cavity so the carboxyl groups were within 3Å of the side chain nitrogen of the αLys99.
CPR was run with a step size of 0.002Å and a required saddle gradient RMS of 0.001Å.
