该研究工作得到了国家高技术研究发展计划(863计划)(No.&&;No.&&)及中国科学院动物研究所知识创新工程(KSCX3-IOZ-0701)课题的资助。
文章通讯作者是伍一军教授,分子毒理学研究组组长。
通过重组DNA技术构建的具有特定功能(如:降解农药)的基因工程,即工程微生物&&(genetically&&engineered&&microorganism,&&GEM)具有广阔的应用前景,然而,GEM的应用本身又有可能引发新的生态问题,这种潜在的危险性反过来限制了它的应用。为了研究如何最小化GEM应用的潜在风险,有必要建立用于环境中GEM的安全监测体系和在需要时能减少或消除其存在的主动生物防御(active&&biologic&&containment,ABC)体系。
动物研究所伍一军研究组利用双质粒表达系统成功构建了具有荧光特性和农药降解活性及可诱导性自杀能力的GEM。
在该双质粒表达系统中有两个功能质粒:一个是用于ABC体系的自杀基因双拷贝质粒;另一个是与自杀质粒相容的带有荧光蛋白基因与农药降解酶基因的功能质粒。前者含有来源于Serratia&&marcescens&&的去除前导肽的核酸酶自杀基因,该基因被构建到T7启动子下游,其表达受阿拉伯糖诱导;后者则是带有有机磷水解酶(organophosphorus&&hydrolase,OPH)基因与绿色荧光蛋白(enhanced&&green&&fluorescent&&protein,EGFP)基因的质粒pL-EGFP-OPH,将其与自杀质粒共转于E.&&coli&&BL21AITM&&中得到GEB。
通过荧光检测和酶活性检测以及可诱导性自杀能力的检测,证明该GEB不仅具有可供方便检测的荧光特性,而且还具有可诱导自杀能力,同时还具有较强的农药降解活性。具有自杀控制功能的安全型GEB的研究成功极大地提高了GEM在环境中实际应用的安全性。
安徽桐城人,博士,佐治亚医学院分子医学与遗传学系基因调控专业博士后。现为中科院动物研究所分子毒理学研究组组长,中科院动物研究所研究员,中科院研究生院兼职教授,博士研究生导师。
分子毒理学研究组多年来一直开展化学杀虫剂与生物大分子的相互作用机理及环境毒理学的研究。承担了国家攻关项目、中国科学院重点项目、国家自然科学基金项目、中科院生物特别支持项目、中科院知识创新项目和国际合作项目,并承担中科院创新重大项目和国家863专题项目以及科技部科技支撑项目和863探索项目。先后开展了对一些重要杀虫药剂和神经毒剂的毒性机理的研究,例如:就溴氰菊酯对脑蛋白质磷酸化的影响开展研究,以了解拟除虫菊酯类杀虫剂的神经毒理机制;对有机磷杀虫剂和其他有机磷引起的迟发性神经毒性(&&OPIDN&&)问题开展研究,探索此类毒剂的神经毒性的分子机制;对生物体中重要的代谢酶系&&――&&细胞色素P450酶系的结构、功能开展研究,为这类酶系在农业和环境中的应用打下基础;对杀虫剂和其它农药使用后的转归开展研究,为农产品及其环境的安全性评价提供依据。在这些研究领域,分子毒理学研究组已经取得了具有较高价值的研究成果,特别是针对OPIDN的研究,跻身该领域国际前沿。业已发现,有机磷杀虫剂引起OPIDN的主要靶标蛋白-神经毒性酯酶(NTE)并非神经细胞分化所必需,但却在神经生长发育过程中起着重要的辅助作用。NTE的过表达可以引起细胞周期阻滞。NTE蛋白可通过泛素蛋白酶体途径降解,也可通过溶酶体途径降解,前者与NTE蛋白的调节域(R-)有关;而后者则与NTE的跨膜结构域和调节域(R-)有关。这些研究成果对于了解NTE的生理功能及其在OPIDN中的作用具有重要的意义。
本研究组先后申请国家发明专利2项,编著出版学术著作2部,在国际核心学术刊物上发表研究数十篇。建立了经过中华人民共和国农业部认证的农药残留分析检测实验室。截至2007年底,分子毒理学研究组已培养研究生15人,其中博士研究生8人,硕士研究生7人;现在读研究生8人,已毕业7人。
目前,分子毒理学研究组最感兴趣的研究方向是:(1)有机磷神经毒性的分子机理:NTE的生理功能及其代谢调节;(2)神经毒性杀虫剂与神经毒性化学物对人畜健康危害的控制以及昆虫与其他生物对杀虫药剂耐受性的机理。(3)环境中有毒物的检测及消解:残留农药的非常规检测技术及利用生物工程技术降解残留农药的环境安全问题。
